I did not see a setting in the Bios to disable the slot but some other setting (or Processor) might effectively do that.
When I was running the 11th Gen on the XII, the PCIe16_1 slot was running at x8. Someone else mentioned they saw that was being done when running PCIe 4..

The M.2_1 and M.2_2 slots both come off the processor with the M.2_1 being disabled with the 10th Gen. .. so it could be processor related..
And there usually aren't any Official ASUS technical folks in these forums.

Can someone from Asus clarify? Why is my PCIE16_1 slot in 8x with just one NVMe plugged in? Please note I am using a 10th gen 10900K but the manual clearly states that 10th gen will run the PCIE16_1 slot in 16x mode.

Might want to try both reseating the CPU and GPU to ensure proper contact.
