I might be mis-identifying it. I am by no means an expert.

As you can see the card has a pair of small male connectors. I thought that little slot type was called a PCI Express.

Yes I believe that you do need to connect it to the small black PCIe connector. all the connectors are PCIe just different bandwiths.
I found this and it may help you to m.2 ssd in asus bios. Enter the bios by pressing del or f2 key when computer boots. Go to advanced and there choose onboard devices configuration. There's pci express x4_3 slot (black) bandwidth, choose m. So you do have to configure that slot for m.2 booting. Seems with my manual there was a procedure for setting this up. there used to be Asus Specific adapters but this may have been as this was all new back then.
