Did you install the drivers on the install in the correct order?

There is something funky going on here. I can make a connection with the PCIe card if it's plugged into a slot that shares bandwidth with the GPU. But the front panel audio uses the SupremeFX chip and the lime connector in the back uses the same old Realtek chip that is several generations old now and both of these run through the *DMI bus down to the PCH controller.*
The only thing at is common to all of these is the PSU and it's totally possible your rails are sagging when the GPU gets loaded. If you have another PSU that would be worth a try. *

Have you used different speakers? Shouldn't make a difference but the rest doesn't make sense either. Maybe your ASUS MOBO got offended when you put a GB graphics card in it?
I cant think of anything else to try that doesn't involve spending more money.

Glad that took care of it but I would be looking for grounding issues with your electrical outlets if that solved the problem.
